C-line IgY IgG DNP

Summary

A C-Line is a control line, which is often used in many different of POCT of IVD industries.  C-line IgY IgG DNP means there is IgY,IgG or DNP under it. If a test line (T) is visible together with a control line (C), this means that the result is positive. Look carefully at the result: The test should be considered positive if two lines are visible. On the other hand, if the C line is not displayed and the C area is white, it indicates that there is a problem with the quality of the test paper used by the patient, so the test paper test results  don't have  reference value.

Animals that produce IgY are basically oviparous, and the main immunoglobulin (IgY) present in the blood of birds is passed on to their offspring and accumulated in the egg yolk. Poultry antibodies are mainly concentrated in the yolk. The antibodies in the yolk account for about 80% of all antibodies in the birds themselves, and the antibodies in the serum account for about 19%. There are three main types of immunoglobulins in poultry, namely IgY, IgM and IgA. IgY is the main immunoglobulin in poultry, and IgY mainly exists in blood and egg yolk.

There is functional homology between avian IgY and mammalian IgG, but there are significant differences in molecular weight, structure and biochemical function.

a. Both are composed of two identical heavy chains and light chains, which are bonded together through disulfide bonds;

b. They have variable antigen-binding sites and constant highly conserved regions.

Differences:

a. IgY has a higher molecular weight, and the molecular weight of IgY is ~167 kDa (heavy chain 65 kDa, light chain 19 kDa); b.IgY heavy chain consists of 4 constant regions (Cv1-Cv4) and 1 variable region; c.IgY does not have a hinge region, while IgG has a hinge structure. This property makes the Fab fragment of IgY hinder the binding of antigens. The special structure of IgY makes it have a larger molecular weight, reduced swing and flexibility, and higher stability;

d. IgY has more hydrophobic groups than IgG, which makes it more suitable for the lipid-rich yolk environment.

Application Prospects

Yolk antibody technology has a wide range of applications in human and veterinary health. It can be used for many types of treatments; it can be used to prevent various types of diseases and can detect a variety of antigens such as microorganisms, tumor markers and substances through different technologies.

The quality of the C-line IgY IgG DNP in POCT detection reagents is crucial. It can make timely, accurate and intuitive judgments on whether the detection reagent product is effective, batch-to-batch consistency and stability, and the accuracy of sample quantification. Currently, the common quality control systems (labeling and coating) in POCT detection reagents include rabbit IgG-goat anti-rabbit IgG, mouse IgG-goat anti-mouse IgG, chicken IgY-goat anti-chicken IgY, and independent quality control DNP-goat anti-DNP. etc.

Chicken IgY-Sheep anti-chicken IgY is more suitable for making C line than other quality control systems:

1. Chicken IgY is easy to obtain, and a large amount of antibodies can be obtained from eggs without blood collection;

2. Mass production of chicken IgY is easy to control, and stable, non-specific antigen-binding antibodies can be obtained from SPF eggs;

3. The possibility of serological cross-reaction between chicken IgY and mammalian immunoglobulin is low because the germline distance      between the two is quite different;

4. Chicken IgY does not activate the mammalian complement system and does not bind to rheumatoid factor or Fc receptors, which can        avoid false negative or false positive results during the immune detection process;

5. Acid-resistant, will not be inactivated at pasteurization temperature and remains stable at pH<3;

6. More stable, convenient and reliable than antibodies extracted from mammals;

7. Mature and stable production technology can ensure the supply of high-quality chicken IgY.
